"Synthesis and characterization of bis (oxime) and bis (dioxime) based transition metal complexes as topoisomerase I and II inhibitors: in vitro DNA binding, nuclease activity and cytotoxicity against various human cancer cells” (2017-2019), funded by The Scientific and Technological Research Council of Turkey (TUBITAK).

In our project, potential bis(oxime) and bis(dioxime) ligands and their coordination compounds with various biological transition metals were synthesized in five different series having different functional groups and their usability as potential anticancer drug was investigated. For this purpose, bis(oxime) and bis(dioxime) ligands were synthesized by reacting with various isonitrosoacetophenone derivatives and chelating aliphatic diamines which will serve as bridges. Coordination compounds with various transition metals such as Cu(II), Ni(II), Co(II) and Zn(II) were isolated. Structures of ligands and complexes have been characterized by using various spectroscopic techniques. DNA binding-cleavage activities were investigated and also binding mode and mechanism of cleavage were determined. Furthermore, the effects of concentration and incubation time for DNA cleavage were studied and optimal conditions were determined. It has been determined that binding mode of metallonucleases with DNA was noncovalent interaction including both of electrostatic and partial groove binding modes together. Also it has been observed that artificial metallonucleases could cleave DNA with both oxidative and hydrolytic pathways and peroxide, singlet oxygen and hydroxyl radicals played role in cleavage. Topoisomerase I and Topoisomerase IIα inhibition activities of ligands and complexes were investigated. Topoisomerase I and IIα inhibition activities of compounds increased depending on the concentration. In general, compounds showed best Topoisomerase I and Topoisomerase IIα inhibitory activity in the complexes were Ni(II) complexes having 1,3- diaminopropane bridge group. Among the bridge groups the best biologically activated ligands were 1,3-diaminopropane, diethylenetriamine and bis(aminoxy)ethane, respectively. One of the conclusions is in this group Ni(II) complexes were the best active metal groups and Cu(II) and Zn(II) complexes follow the order. Among the Ni(II) complexes, best binding activity was found to be during complexes containing -OCH3, -naphthyl, -Cl, -CH3 and -benzene. In anticancer activity studies, another important result obtained that early apoptosis was %98 from all doses at the [Cu3(L9)2]Cl2 complex.
